Hello friends, And viruses, an organism whose name reminds us of diseases, the same viruses are being used by scientists to cure a deadly disease like cancer. And you know what? Many human experiments of this technology have been successful.

Recently, cancer surgeon Yuman Fong, with the help of these viruses, has also succeeded in curing the last-stage cancer patient. At a stage when it was believed that cancer could not be cured. In fact, according to doctors, these viruses can cure more than 8 cancers like breast, lung, and ovarian.

So, can we make cancer a 100% curable disease with this technology? Well, actually, let's not jump the guns. Because, in the 1950s, when the cancer-curing abilities of the virus began to be tested, many patients died. Actually, this process started in 1904, where a patient of leukemia, i.e. blood cancer, was infected with the influenza virus.

During his treatment, an American doctor, George Dock, saw a very strange thing. Because of this viral infection, his cancer was being cured. Because of cancer, his liver and spleen, which had swollen, were returning to their normal size again. And even these organs were becoming cancer-free.

Now, how was this viral infection killing cancer cells? George tried to find out this mechanism through many experiments. But, because of the lack of technology, he could not do anything at that time. And so, in the end, he published all the observations made in this patient only through a paper. So that in the future, if there is any study on this, the actual reasons behind it can be revealed to everyone.

Now, a few years later, research on this begins. And this is seen by the famous oncologist, i.e. cancer specialist, Chester Southam. So, he thought of moving the study forward. Now, he knew that the virus was the reason behind it. And so, to move the study forward, he started injecting different viruses into cancer patients and studying its effects.

In 1956, the first trial was started and first of all, he injected the common flu virus, adenovirus, into 30 cancer patients and observed the cancer. He saw that the size of the cancer cells, i.e. tumors, of 26 out of 30 patients were slowly decreasing. And they were recovering. Seeing the success of the first trial, he started the second trial. And this time, he infected 22 cancer patients with the second virus, Hepatitis B. And again, he observed them.

Now, in the beginning, the results of this experiment were the same. But within a few days, 14 out of 22 patients started showing severe symptoms of Hepatitis B. Their skin started turning yellow, they started having fever, and abdominal pain kept increasing, and unfortunately, one patient lost his life. But friends, this is not the end of the story.

On the other hand, as soon as the viral infection of the rest of the patients recovered, their tumor size started increasing again, at twice the rate. And as soon as he saw it, in the interval of one to two months, some of the patients died. Now, this was a very strange thing, right? But nevertheless, after seeing such shocking results of this study, this study was immediately banned. And there, his research remained incomplete.

Until 1990, when a new branch of the medical field, Genetic Engineering, started, which changed the whole world of medicine. Basically, in genetic engineering, with the help of technology, genes are modified by adding or cutting them, so that diseases can be treated and cured.

Now, this process was showing a lot of positive results. And that is why it was used to make the world's first genetically modified vaccine, Hepatitis B. To make this vaccine, scientists extracted HBS antigen from yeast. Now, specifically HBS antigen here, because this same antigen is also present on the surface of the Hepatitis B virus.

And that is why they thought that by using this antigen, they can identify the Hepatitis B virus in the body and destroy it. How? So see, whenever an antigen, that is a foreign particle, enters the body, then to kill it, the human immune system produces specific antibodies which only attack those antigens.

That is, because of HBS antigen, the antibodies produced cannot attack the antigens of any other disease. So they introduced this antigen from the body through the vaccine so that when the Hepatitis B virus enters the body, this antigen identifies the HBS antigen present on its surface and produces those specific antibodies and along with the antigen, kills the entire virus.

With the help of this vaccine, doctors were able to bring Hepatitis B under control. Now, looking at the successful results of this vaccine, in 1996, cancer biologist Frank McCormick and his team thought that in the same way, by changing the genes of the virus, the technique of cancer treatment can be revived.

And that's why, just like in 1956, they took the Adenovirus. But this time, with the help of genetic engineering, they made some changes in its genes and made a new virus called Onyx 015. Now, this specific modification proved to be a game changer.

See friends, Protein P53 is attached to our cells and works like a tumor suppressor on them. This means that it regulates cell division and also sends a signal to be destroyed at the right time. But in cancer cells, this protein gets deactivated because of which cells do not die over time and keep making their copies.

And in this Onyx 015, modifications were made to target these cells so that the virus targets only those cells in which protein P53 was deactivated. So after these modifications, they started its lab tests. And for this, they first took normal cells in the lab and exposed them to radiation to turn them into tumor cells.

And then they injected that virus and observed it for a few days. After a few days, they saw that Onyx 015 ignored normal cells and was only targeting cancer cells and killing them. But here, after a few more observations, they saw a problem in the results.

According to their results, Onyx 015 was not able to replicate itself quickly and also the modifications made to its genes were not passing on to the next generation. So basically, what happens is that any organism's packaging capacity has a limit to accepting new genes.

On top of that, it neither accepts new genes nor passes them on to the next generation. And the same thing was happening with Onyx 015. Because of which, the next generation of the virus was not only killing cancer cells but also healthy cells.

But, to solve this problem, they came up with a solution. They started modifying a virus that was shockingly present in our bodies since childhood. So, to avoid the problems that were emerging in Onyx 015, some other scientists thought to take a virus that our human body already knows and has no side effects.

When they were researching this, they thought of a viral disease that had become extinct. Smallpox. Vaccinia is caused by the virus.

Now, specifically, smallpox is there because we are already vaccinated against this disease. So, even if it is injected into the body, there is no chance of any kind of infection. So, leading this second study, Professor Yumen Fong considered Vaccinia and started doing genetic modifications like Onyx 015.

He noticed that this virus can accommodate many new genes at the same time. Because of this, it was easier to make it cancer-specific. Because normally, as soon as the virus enters the body, it goes inside the cell to hide. So, our immune system cannot detect it.

The virus activates in this cell and quickly starts making a lot of copies of itself. As a result, after a point, there are so many copies of the virus in the cell that it cannot be adjusted and bursts, which is called a viral load. Now, when the cell bursts, these copies go into the surrounding cells and follow the same process, which causes the cells to die and the virus to multiply.

Seeing the increasing number of this virus, the immune system of the body starts making antibodies. These antibodies then attack the virus and kill it, which increases the body's temperature in response. Now, to prevent all this, this virus was modified in such a way that it could not activate the immune system of the human body. In fact, such genes were added that would produce proteins whose only function would be to kill only cancer cells.

So, this process was set up in such a way that as soon as the vaccine or virus enters the human body, it will first produce Max Protein. This Max Protein will detect the MYC protein present on the surface so that the virus can only enter those cells and have no effect on the rest of the normal cells. Then, as soon as the cancer cell is detected, the virus will enter that cell and produce its second protein, Thymidine kinase, which will convert the non-toxins present in the cells into cytotoxins.

These cytotoxins are generally present in snake venom, which are diffused in the cells and force them to commit suicide. Now, when this process starts, the virus will release its third and final protein, Thrombospondin. This protein will increase the angiolysis process, due to which the blood vessels will be damaged and the blood supply to the cancer cells will stop.

As a result, their functioning will stop. Now, whether this whole process will be successful or not, we will get to know about the special protein that is already present in the vaccine. HNIS means Human Sodium Iodide Simporter.

Well, this protein only accepts Iodine in the cancer cells. In short, HNIS works like a security system that allows specific elements, in this case, Iodine, to enter the cells. In this way, when radioactive Iodine is injected during tumor detection, it is only accepted in the place where the cancer cells are present, which makes it easier to track them.

Along with this, it also helps in radiotherapy, in which the tumor is destroyed by X-rays. So, when the scientists believed that the vaccine was giving them the expected results, they started testing it. For this, first of all, they conducted animal trials, in which rats, guinea pigs, and monkeys were included.

Keeping in mind the good results of these trials, FDA gave permission for its human trial. And in 2022, the US City of Hope Hospital and Australian company ImmuGene together started the trials of Phase 1 of this therapy, where for the first time, this virus was injected into humans. Initially, they selected 100 such patients who were suffering from Solid Tumor, i.e. Advanced Cancer.

For their safety, they were given an extra drug, Pembrolizumab, along with the virus. In Phase 1 trials, they saw that patients' pancreatic, lung, breast, ovarian—more than eight types of tumors—were decreasing, i.e. they were proving successful. Now, seeing these results, the US doctors are also thinking to start Phase 2 and 3 trials soon, which will be conducted on around 10,000 to 50,000 patients.

For now, this will be limited to the US only. But similarly, another trial is being conducted here in Mumbai, where Tata Memorial, ACTRAC, and IIT Bombay are working together. This treatment is a little different from Vaccinia because in this, the virus is fused with the T-cells, and a drug is made, which is also called Chimera or CAR T-cell therapy.

Now, because some cells of our immune system are included, this drug will attack the cancer cells aggressively. The main motive behind making this drug in India was to make cancer treatment affordable for the common people. Because in many other countries, the price of this drug and treatment goes up to 5 to 10 crores. And in India, its treatment will cost around 20 lakhs.
